Tafinlar 50 mg Capsules Tafinlar contains dabrafenib, an oral kinase inhibitor targeting mutated BRAF serine/threonine kinases. Specific BRAF gene mutations, such as BRAF V600E and V600K, lead to constitutive activation of the BRAF pathway, driving downstream MEK and ERK signaling to promote oncogenic cellular proliferation and survival. Dabrafenib selectively inhibits BRAF V600 mutation-positive kinase activity, suppressing cell proliferation and inducing apoptosis in BRAF-mutated tumor cell lines. Tafinlar is indicated, either as monotherapy or in combination with trametinib (Mekinist), for the treatment of patients with unresectable or metastatic melanoma with a BRAF V600 mutation, metastatic non-small cell lung cancer (NSCLC) with a BRAF V600E mutation, and BRAF V600E mutation-positive anaplastic thyroid cancer (ATC) or solid tumors.
