Tivdak 40 mg injection Tivdak contains tisotumab vedotin-tftv, an antibody-drug conjugate (ADC) composed of a human monoclonal antibody directed against tissue factor (TF) covalently linked to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a protease-cleavable linker. Tissue factor is aberrantly expressed on various solid tumor cells, playing a role in tumor growth, angiogenesis, and metastasis. Upon binding to tissue factor on the surface of tumor cells, the ADC complex is internalized. Intracellular lysosomal proteases cleave the linker, releasing active MMAE. MMAE binds to tubulin and disrupts the microtubule network within the cell, leading to cell cycle arrest and apoptotic cell death. Tivdak is indicated for the treatment of adult patients with recurrent or metastatic cervical cancer with disease progression on or after chemotherapy.
